He doesn't say "Roy," he says "Roid" (or "Royd"). That'd be short for "hemorrhoid," which is a very literal pain in the butt. Google it if you want, although you may be in for some serious TMI.
So he's making a pun off of Boyd's name and the fact that Boyd's being a pain in the butt, in other words.
